    Geochronological challenges posed by continuously developing tectonometamorphic systems: insights from Rb–Sr mica ages from the Cycladic Blueschist Belt, Syros (Greece).

    Is metamorphism and its causative tectonics best viewed as a series of punctuated events or as a continuum? This question is addressed through examination of the timing of exhumation of the Cycladic Blueschist Belt (CBB). The cause of scatter beyond analytical error in Rb–Sr geochronology was investigated using a suite of 39 phengite samples. Rb–Sr ages have been measured on phengite microsamples drilled from specific microstructures in thin sections of calcschists and metabasites from the CBB on Syros. The majority are from samples that have well-preserved blueschist facies mineral assemblages with limited greenschist facies overprint. The peak metamorphic temperatures involved are below the closure temperature for white mica so that crystallization ages are expected to be preserved. This is supported by the coexistence of different ages in microstructures of different relative age; in one sample phengite from the dominant extensional blueschist facies fabric preserves an age of 35 Ma while post-tectonic mica, millimetres away, has an age of 26 Ma. The results suggest that micro-sampling techniques linked to detailed microstructural analysis are critical to understanding the timing and duration of deformation in tectonometamorphic systems. North of the Serpentinite Belt in northern Syros, phengite Rb–Sr ages are generally between 53 and 46 Ma, comparable to previous dates from this area. South of the Serpentinite Belt phengite in blueschist facies assemblages associated with extensional fabrics linked to exhumation have ages that range from 42 Ma down to c. 30 Ma indicating that extensional deformation while still under blueschist facies conditions continued until 30 Ma. No age measurements on samples with unambiguous evidence of deformation under greenschist facies conditions were made; two rocks with greenschist facies assemblages gave phengite ages that overlap with the younger blueschist samples, suggesting blueschist facies phengite is preserved in these rocks. Two samples yielded ages below 27 Ma; one is from a post-tectonic microstructure, the other from a greenschist in which the fabric developed during earlier blueschist facies conditions. These ages are consistent with previous evidence of greenschist facies conditions from c. 25 Ma onwards. The data are consistent with a model of deformation that is continuous on a regional scale

    Phosphorus dynamics in the Barents Sea

    The Barents Sea is considered a warming hotspot in the Arctic; elevated sea surface temperatures have been accompanied with increased inïŹ‚ow of Atlantic water onto the shelf sea. Such hydrodynamic changes and a concomitant reduction of sea ice coverage enables a prolonged phytoplankton growing season, which will inevitably affect nutrient stoichiometry and the controls on primary production. During the summer of 2018, we investigated the role of phosphorus in mediating primary production in the Barents Sea. Dissolved inorganic phosphorus (DIP), its most bioavailable form, had an average net turnover time of 9.4ïżœ4.8 d. The most southern Atlantic inïŹ‚uenced station accounted for both the highest rates of primary production (655 mg C m2 d−1) and shortest net DIP turnover (2.8ïżœ0.5 d). The fraction of assimilated DIP released as dissolved organic phosphorus (DOP) at this station was < 4% compared to an average of 21% at all other stations. We observed signiïŹcant differences between phytoplankton communities in Arctic and Atlantic waters within the Barents Sea. Slower DIP turnover and greater release of DOP was associated with Phaeocystis pouchetii dominated communities in Arctic waters. Faster turnover rates and greater phosphorus retention occurred among the Atlantic phytoplankton communities dominated by Emiliania huxleyi. TheseïŹndings provide baseline measurements of P utilization in the Barents Sea, and suggest increased Atlantic intrusion of this region could be accompanied by more rapid DIP turnover, possibly leading to future P limitation (rather than N limitation) on primary productio

    Rnf165/Ark2C enhances BMP-smad signaling to mediate motor axon extension

    Little is known about extrinsic signals required for the advancement of motor neuron (MN) axons, which extend over long distances in the periphery to form precise connections with target muscles. Here we present that Rnf165 (Arkadia-like; Arkadia2; Ark2C) is expressed specifically in the nervous system and that its loss in mice causes motor innervation defects that originate during development and lead to wasting and death before weaning. The defects range from severe reduction of motor axon extension as observed in the dorsal forelimb to shortening of presynaptic branches of the phrenic nerve, as observed in the diaphragm. Molecular functional analysis showed that in the context of the spinal cord Ark2C enhances transcriptional responses of the Smad1/5/8 effectors, which are activated (phosphorylated) downstream of Bone Morphogenetic Protein (BMP) signals. Consistent with Ark2C-modulated BMP signaling influencing motor axons, motor pools in the spinal cord were found to harbor phosphorylated Smad1/5/8 (pSmad) and treatment of primary MN with BMP inhibitor diminished axon length. In addition, genetic reduction of BMP-Smad signaling in Ark2C+/− mice caused the emergence of Ark2C−/−-like dorsal forelimb innervation deficits confirming that enhancement of BMP-Smad responses by Ark2C mediates efficient innervation. Together the above data reveal an involvement of BMP-Smad signaling in motor axon advancement

    Solid-Phase Microextraction and the Human Fecal VOC Metabolome

    The diagnostic potential and health implications of volatile organic compounds (VOCs) present in human feces has begun to receive considerable attention. Headspace solid-phase microextraction (SPME) has greatly facilitated the isolation and analysis of VOCs from human feces. Pioneering human fecal VOC metabolomic investigations have utilized a single SPME fiber type for analyte extraction and analysis. However, we hypothesized that the multifarious nature of metabolites present in human feces dictates the use of several diverse SPME fiber coatings for more comprehensive metabolomic coverage. We report here an evaluation of eight different commercially available SPME fibers, in combination with both GC-MS and GC-FID, and identify the 50/30 ”m CAR-DVB-PDMS, 85 ”m CAR-PDMS, 65 ”m DVB-PDMS, 7 ”m PDMS, and 60 ”m PEG SPME fibers as a minimal set of fibers appropriate for human fecal VOC metabolomics, collectively isolating approximately 90% of the total metabolites obtained when using all eight fibers. We also evaluate the effect of extraction duration on metabolite isolation and illustrate that ex vivo enteric microbial fermentation has no effect on metabolite composition during prolonged extractions if the SPME is performed as described herein

    The personal experience of parenting a child with Juvenile Huntington’s Disease: perceptions across Europe

    The study reported here presents a detailed description of what it is like to parent a child with juvenile Huntington’s disease in families across four European countries. Its primary aim was to develop and extend findings from a previous UK study. The study recruited parents from four European countries: Holland, Italy, Poland and Sweden,. A secondary aim was to see the extent to which the findings from the UK study were repeated across Europe and the degree of commonality or divergence across the different countries. Fourteen parents who were the primary caregiver took part in a semistructured interview. These were analyzed using an established qualitative methodology, interpretative phenomenological analysis. Five analytic themes were derived from the analysis: the early signs of something wrong; parental understanding of juvenile Huntington’s disease; living with the disease; other people’s knowledge and understanding; and need for support. These are discussed in light of the considerable convergence between the experiences of families in the United Kingdom and elsewhere in Europe

    Exoplanets and SETI

    The discovery of exoplanets has both focused and expanded the search for extraterrestrial intelligence. The consideration of Earth as an exoplanet, the knowledge of the orbital parameters of individual exoplanets, and our new understanding of the prevalence of exoplanets throughout the galaxy have all altered the search strategies of communication SETI efforts, by inspiring new "Schelling points" (i.e. optimal search strategies for beacons). Future efforts to characterize individual planets photometrically and spectroscopically, with imaging and via transit, will also allow for searches for a variety of technosignatures on their surfaces, in their atmospheres, and in orbit around them.     Allellic variants in regulatory regions of cyclooxygenase-2: association with advanced colorectal adenoma

    Cyclooxygenase 2 (Cox-2) is upregulated in colorectal adenomas and carcinomas. Polymorphisms in the Cox-2 gene may influence its function and/or its expression and may modify the protective effect of nonsteroidal anti-inflammatory drugs (NSAIDs), thereby impacting individuals' risk of developing colorectal cancer and response to prevention/intervention strategies. In a nested case–control study, four polymorphisms in the Cox-2 gene (two in the promoter, −663 insertion/deletion, GT/(GT) and −798 A/G; one in intron 5-5229, T/G; one in 3â€Čuntranslated region (UTR)-8494, T/C) were genotyped in 726 cases of colorectal adenomas and 729 age- and gender-matched controls in the prostate, lung, colorectal, and ovarian (PLCO) cancer screening trial. There was no significant association between the Cox-2 polymorphisms and adenoma development in the overall population. However, in males, the relatively rare heterozygous genotype GT/(GT) at −663 in the promoter and the variant homozygous genotype G/G at intron 5-5229 appeared to have inverse associations (odds ratio (OR)=0.59, confidence interval (CI): 0.34–1.02 and OR=0.48, CI: 0.24–0.99, respectively), whereas the heterozygous genotype T/C at 3â€ČUTR-8494 had a positive association (OR=1.31, CI: 1.01–1.71) with adenoma development. Furthermore, the haplotype carrying the risk-conferring 3â€ČUTR-8494 variant was associated with a 35% increase in the odds for adenoma incidence in males (OR=1.35, CI: 1.07–1.70), but the one with a risk allele at 3â€ČUTR-8494 and a protective allele at intron 5-5229 had no effect on adenoma development (OR=0.85, CI: 0.66–1.09). Gender-related differences in adenoma risk were also noted with tobacco usage and protective effects of NSAIDs. Our analysis underscores the significance of the overall allelic architecture of Cox-2 as an important determinant for risk assessment

    Focal brain trauma in the cryogenic lesion model in mice

    The method to induce unilateral cryogenic lesions was first described in 1958 by Klatzo. We describe here an adaptation of this model that allows reliable measurement of lesion volume and vasogenic edema by 2, 3, 5-triphenyltetrazolium chloride-staining and Evans blue extravasation in mice. A copper or aluminium cylinder with a tip diameter of 2.5 mm is cooled with liquid nitrogen and placed on the exposed skull bone over the parietal cortex (coordinates from bregma: 1.5 mm posterior, 1.5 mm lateral). The tip diameter and the contact time between the tip and the parietal skull determine the extent of cryolesion. Due to an early damage of the blood brain barrier, the cryogenic cortical injury is characterized by vasogenic edema, marked brain swelling, and inflammation. The lesion grows during the first 24 hours, a process involving complex interactions between endothelial cells, immune cells, cerebral blood flow, and the intracranial pressure. These contribute substantially to the damage from the initial injury. The major advantage of the cryogenic lesion model is the circumscribed and highly reproducible lesion size and location

    Upregulation of P2Y2 receptors by retinoids in normal human epidermal keratinocytes

    Retinoids, vitamin A derivatives, are important regulators of the growth and differentiation of skin cells. Although retinoids are therapeutically used for several skin ailments, little is known about their effects on P2 receptors, known to be involved in various functions in the skin. DNA array analysis showed that treatment of normal human epidermal keratinocytes (NHEKs) with all-trans-retinoic acid (ATRA), an agonist to RAR (retinoic acid receptor), enhanced the expression of mRNA for the P2Y2 receptor, a metabotropic P2 receptor that is known to be involved in the proliferation of the epidermis. The expression of other P2 receptors in NHEKs was not affected by ATRA. ATRA increased the mRNA for the P2Y2 receptor in a concentration-dependent fashion (1 nM to 1 ΌM). Am80, a synthesized agonist to RAR, showed a similar enhancement, whereas 9-cis-retinoic acid (9-cisRA), an agonist to RXR (retinoid X receptor), enhanced P2Y2 gene expression to a lesser extent. Ca2+ imaging analysis showed that ATRA also increased the function of P2Y2 receptors in NHEKs. Retinoids are known to enhance the turnover of the epidermis by increasing both proliferation and terminal differentiation. The DNA microarray analysis also revealed that ATRA upregulates various genes involved in the differentiation of NHEKs. Our present results suggest that retinoids, at least in part, exert their proliferative effects by upregulating P2Y2 receptors in NHEKs. This effect of retinoids may be closely related to their therapeutic effect against various ailments or aging events in skins such as over-keratinization, pigmentation and re-modeling
